There are 5 harlekins in the city, and you have to beat them with your fists. If you you accidentally kill one, use a fast travel tunnel, then he should be there again.
They are all near the water, four in the centro district, one of them is under a bridge,
and the one in the campagne district is near the house

The five Harlequins are found throughout the Campagna (1) and Centro (4) districts. As the video and others say, the Harlequins do glitch in and out, so go to a tunnel and return to the area. DO NOT KILL, only beat them up with your fists.

Harlequin Locations
Note: Sometimes they won't appear so run away and come back. Or use a Tunnel to travel away and then back.
Also, if they die by some other means of your fists, do the same thing as above. They respawn.
You have to beat up (use must use your fists) 5 thirsty Harlequins. You will know if it counted because it says "X of 5 Harlequins beaten up." They are near the water, hence the thirsty part. So look along the river. 4 are in Centro District and 1 is in Campagna District.

I. The First Harlequin is in Centro District North and a bit west beside the river. He is between the Highest Doctor symbol on the map and the second Highest Tunnel Entrance. He is on some stairs going into the water.
II. The Second Harlequin is in Centro District North West beside the river again. He is to the East of the Bridge that goes to Vanticao District. Starting from the beginning of the bridge, if you use your map and go along the river East you will see a shaded area with a lighter circle in the middle. Go North of that circle and you will see a set of double stairs. He is on those stairs.
III. The Third Harlequin is in Centro District West beside the river. Go to ROSA IN FIORE and go directly west from there. He is on a set of stairs again.
IV. The Fourth Harlequin is in Centro District South East under a bridge beside the river. Go to Ezio's Hideout and go DIRECTLY south of it. On your map you should see a Bridge that has lines going across it. He is under that bridge on the left side.
V. The Fifth and Last Harlequin is in Campagna District near the Borgia Tower (Or Assassin's Tower) that is surrounded by water and cliffs. If you use your map and go West slightly from the Borgia/Assassin's Tower and then go slightly North, you will see a small rectangle that represents a small building. Go there and the Harlequin is behind the Small building along the edge of the cliff. |

| Can confirm that if one doesnt spawn, go kill some guards (by your own hand, no help) I killed 4, went back with a fist and he was there. also, you can walk away slowly and they will follow you away from the water. Under the bridge is useful for that. |

A friend recommended I put some of my AC Brotherhood walkthroughs on this site. So, here's a video walkthrough I made myself of how to get "Clowning Around."
The Harlequins don't actually glitch (I've seen that mentioned on this site,) they just appear randomly. That's how they're supposed to be. And if you knock them into water, they do respawn, but not necessarily immediately.

Additional info: Easiest way to knock them out cold & avoid them to land in the water...
target the harlequin, walk towards him and grab him with B... throw him away from the water or against a wall or something... go stand behind him, wait till he gets back up and push X, an animation will show up of you executing the harlequin
(must have bare fists, no hidden blade!) |

The other guides were helpful in locating the Harlequins, but it was a challenge to keep them from falling into the water while fighting.
Solution: I simply threw a Smoke Bomb, switched to fists, and hit X for an execution. Very quick and no worries about water. |

| If the harlequin won't appear, I went and bumped into someone so they dropped their lantern and yelled at me. Then the guy appeared. This was repeatable. |
