If you decide to choose Human mode for your first playthrough, then you should obtain it near the end of your second playthrough (on Devil Hunter Mode). However, to get this slightly quicker, complete your first playthrough on Devil Hunter mode to unlock Bloody Palace, where you obtain kills a lot faster than you would going through the story mode.

Also helps when going for the 10,000 kills achievement.
