This popped for me in Level 1, in the panel which starts with Wolverine mentioning his Hold ability. The moment that it popped was when I destroyed a large, dark-looking metal box at the top left corner of the panel, which you get to after going left when you see a 2nd set of three enemies in a line.

I did restart this panel a couple of times, as I thought that I had made a mistake in where I was supposed to be going, and each time I restarted this panel, I destroyed all of the boxes, giving me more Hero Points 'in a single panel'.

Edit: The user "blackboxrory" has confirmed that you can restart the panel and the 'count' towards the achievement will not get reset.
