The easiest way by far to get this achievement (and the other Boss cheevos) is to be on the Easy action difficulty as the Bosses are pushovers.
You fight the Missionary about halfway through the game, right after finding your way back to your apartment. Make sure you have your Shotgun equipped and ready (the Shotgun is found in the Subway, don't think you can miss it). As soon as the fight starts, start unloading your Shotgun onto the Missionary. It may run off but don't worry; just chase after it to catch its attention again. It should only take about 5 Shotgun blasts to down it. Once it is down, run over to it and kick it a few times to finish it off. You really can't fail getting this one. |
