quinnpm already has a great solution to this but if you want to get this on ur 1st playthrough on normal with dante before mission 20 equip one of ur guns as Kalina Ann. Stand in middle during the credits and shoot quickly. All common hell pride will die in one shot getting you alot of quick kills. Any other will take alot of shots so if you want to kill them use ur swords if no other enemies spawn. Your rockets have a range limit so any that are at the edges you will need to move closer to be able to hit them. At 99 kills a hell vanguard will spawn (boss from mission 2) use ur swords to kill him as soon as possible for ur 100th kill. More hell guards will spawn after he has died incase you are not at 100 yet. Kills by lady will not count towards your total. Also i had not leveled up any of my guns so level 1 will be enough to get this out of the way. If you have trouble getting it this way wait until later playthroughs and use quinnpms method.
