Further to Kun's solution, you'll get many of the required kills just playing through the Prologue and Chapter 1.
After that, the quickest way to reach 500 in Chapter 2 is to grind harpies outside Visionary's hut North North East of the Kaedweni Camp. The hut will be marked on your map after you finish the Lost Lambs quest.
When you reach the hut infinite harpies will spawn until you near the line of candles near the front door. In my experience, the line that stops the infinite spawn is about 1-2 metres OUTSIDE the circle of candles. The infinite harpies stopped spawning for me when I got close to the candles even though I didn't cross them. Which is a shame, because with the Blasphemer oufit (which is available only on Dark difficulty) you can hack away endlessly with virtually no chance of dying.
Once you can't maintain the infinite spawn any longer, cross the line to end the infinite spawn. After that, you can spawn more harpies by moving backward and forward between the candle circle and the ravine you came out of. You'll either trigger 4 harpies at once, or 2 followed by 2 more. Once those 4 are dead, you can spawn 4 more by running back and forth again.
Using this method, I was killing 4-8 harpies per minute. Even allowing for breaks to run to the trader and sell/store loot, that'll get you at least 200 kills in an hour and that should be enough to ding this achievement.
As an aside, don't sell all of your harpy feathers. You'll need a total of 72 for the Sackful of Fluff quest. |

You will pretty much get this achievement during your walkthrough. But if you want to get this achievement sooner than later... there are few places where you can "grind" this one.
If you choose Roche's side, you will have a Sackful of Fluff quest during Chapter 2. On this quest a "Collector" wants you to collect Harpy's feathers. Near his cabin there will be a LOT of these enemies. If you meditate 1-2 days forward they will respawn again, during nights there will be even more of them.
It's quite easy to grind this achievement on this area while you are doing this quest and you can always meditate and get them to respawn again.
Edit: You get Sackful of Fluff quest also on Iorveth's side and sooner than on Roche's side. And like o0ArA0o said on comments, you can also grind kills near Visionary's hut, Northeast of the Kaedwen camp.
