This can first be done on the second part of the first level. Use the left trigger for rockets. You will only have three form the start. More can be picked up thought the level. As for total kills there are a couple more parts in other levels to get more. Kills do count even if you die. You will also be working towards the Road Hog and Own The Road achievements for killing bikers.
