For this challenge, I went to the warehouse in Little Italy (I forget the name, but I know it's not Verona) and would clear out the warehouse of mobsters and then kill myself and repeat. You should get about 25 or so mobsters per run and should take about 2-3 minutes per run. Try to avoid mob wars as it tends to slow things down drastically. Street Sweeper and 45+ Respect Level recommended.

I used the Tattaglia Compound in Brooklyn (northeast). Clear out as many as you can with the Street Sweeper. You'll spawn just around the corner from the Compound when you die.

I actually managed to get about 45 kills in one run using this. You'll finish with tons of time left over.
