I used a leveled up Big Man with a baseball bat using "Skull Crush" to get this one. (Sounds like the game of "Clue")

I hired the stock character (since he leveled up automatically) on the second to last mission (mostly, because I forgot to set it up earlier and I had a saved game)

I used Looney (with knife), Daredevil, Big Man, and my main guy (with Tommy Gun).

In the first room, there were only two guys, so I set up a scenario where I could make an opponent "vulnerable" to help Big Man's chance to hit.

A Vulnerable enemy has a +10% chance to hit, so Big Man (with aiming - LT (left trigger)) was around 70% - still took me three "Skull Crush" swings to get the 100 damage points (1 miss, 89 points, then 106 points)

The three "talents" is used to get vulnerability were (I do not believe these abilities stack - so you really only need one):
Suppression Fire (from Tommy Gun)
Slice (from Knife)
Spot Target (character Talent - Daredevil in my case)

Note: There are multiple ways to "respec" Big Man to improve his chances at both hitting and damage, but this method was fast enough for me and it was during a mop up run, so nothing was on the line in terms of story related gaming.
