I recommend attempting this achievement on the "Catacombs" Map (available for download on the Xbox Live Marketplace for 80MSP/$1.00 USD) with 1-5 other players. We attempted with 3 players and found it to be quite fast. The previous solution said it could take up to 4 hours to attain this achievement, but we did it in less than 3. As stated in previous solutions, all creature kills (not just player-controlled) count towards the 100 creature kills and can be collected quickly with just one other person.

Take turns being the odd man out (2-5 players on Alpha Team and 1 on Bravo Team). The creature player turns the crank that operates the floor spikes located in the corridor with a Whopper spawn (big, fat zombie). The player-controlled agents run towards the spikes and slide into them (Hold A and press/hold LT). Sliding prevents you from being knocked back. Keep your agents laying in the spikes after being incapacitated and they will quickly net the creature player a kill. This yields anywhere from 8-13 agent kills per 30-45 sec. round, per person. The second round of each match could be done the same way, or someone could quickly kill the AI-controlled BSAA Agent "Rookie" to count towards his/her "Civilian Casualties" achievement.

If you have a large party (more than 3) you can have one person protect the BSAA Agent "Rookie" to keep the round going as long as possible. If you only have three players like we did it is just faster to have all available agents running towards the spikes.
