I have just got this achievement and you need to do it on Story Mode. Also you must make sure all EXTRAS are switched off and that the final blow is from Batman (Not Robin)

The levels you need to do are

Stage 1-5 - Two Face and Riddler
Stage 2-1 - Catwoman
Stage 3-5 - The Joker
