This achievement pops as soon as you hit the kill count. There is probably a total of around 30 to 35 enemies to shoot in total so you will have to be pretty quick on the draw. After leaving town on the coach you will still have a couple more (about 5) enemies to shoot that will chase you down. Once they start throwing dynamite, your chances are over and if you haven't gotten it, you should reload.

Just remember to get good shots with your guns (pistols recommended) on the men on horseback and you should get this achievement in no time
