You have to defend Cereza(the little girl) during Chapter 6 "The Gates of Paradise". The easiest way to do this is to lure all of the enemies away from Cereza and kill them quickly. They will not go for Cereza as long as they're away from her. Also, if you have 2-3 Golden Flutes, you can use them on every wave to make it easier.
