Best to do on Amateur difficulty with the Majini being easier to kill. There are easily enough of them, you will just have to be patient and wait around a while in each section for all of the Majini to spawn. I personally got the achievement right before heading through the door into the last section.
Do the AI's kills add up as well?
I would assume not with the description stating 'singlehandedly' but I may be wrong. There is a way to stop the AI stealing your kills; go into the AI's inventory (Y then RB) and then select the AI's gun and request it. The AI will still be able to knife enemies but rarely will they be able to kill them.

Also of note: the achievement will unlock as soon as you reach the killcount, so you don't have to wait until the end-screen to see if you achieved the target.Note - You could incorporate this with the 'Shoot the Messenger' achievement
