The best location to do this one is the gather at the Diner. Set up a lot of river traps and buy some more if you feel the need from the vendor machine near by. Once the Little Sister starts collecting the ADAM, head back and hide behind the news TV (it should be in the opposite direction to the entry of the Diner. Watch out for any incoming Splicers from the sides and you should get it.
