You'll run into Harley several times in the game. You'll run into her when you have to save the two guards hanging over the electrified water, and then you'll see her when she's in a guard tower. You'll defeat several waves of enemies on an electrified floor ,and then a cut scene will occur where she tries to take on Batman (She tries and fails.)

After Batman locks her up in a cell, the achievement will pop afterwards.

Story related and cannot be missed.
