You will have to defeat 250 enemies in a game to get credit. If you die, immediately press A to continue and you points will rollover. PLEASE NOTE: If you die on a multiplayer game, it is very possible to lose your points by not pressing continue quickly.
Best suggestion: Play game on normal, keep game continuing and monitor your point score in the top of the screen. Personally I earned the achievement in one Japanese version game before I got to last battles with Magneto.

Just playing through the game and spamming your mutant powers on large groups when available. I got my achievement about 3/4 through the game.
Note: For some reason, it did not unlock for the second person when playing local multiplayer. |

This one I earned with Colossus.
I recommend playing on Expert and on the Japanese version, since more enemies would come out. In each section of the stage, where the enemies come in, make sure to lure them near each other, then go ahead and press Y and you should earn a lot of points for those kills with Colossus's super-power.
You do this right and by halfway through the third stage, where you falling from that trap and then hitting those sand monsters with your super-power, you should have earned 250 points. I earned EXACTLY that when I got the achievement. It's that simple! Should take at least 15-20 minutes out of your time, but it's worth it in the end.
