The best place for this is in the Vaticano District, as there are always plenty of guards around.

Make sure you've got some parachutes in your Inventory, or buy some from a Tailor if needed.

Cross the bridge from the Centro District to the Vaticano, and walk through the gates of the Castel Sant'Angelo (the first building as soon as you enter the Vatican). Climb up the wall surrounding the building, nearest the riverside. As this is a restricted area, you'll probably get chased by some guards but just ignore them. Once up on the wall, run to the furthest West you can and on to the roof on the left hand side. As a reference point, when on that roof you'll be in the corner of the red square marking the restricted area on your map.

From the roof, look down to the non-restricted area and you should see four guards huddled close together in the left corner. Jump off the roof, press X to open your parachute and glide your way towards them. Press X again when you it says Assassinate in the top right HUD, and because of how close they are all stood together you should easily get two of them.
