    ***************SPOILER ALERT****************

This achievement cannot be missed, as it's part of the story.
When you get to chapter 6, you will eventually come across GlaDOS in her potato form. She will ask you to carry her so you can both stop Weathley. Once you pick her up, a switch under her will activate a bridge and the achievement will pop.

P.S. this part is when you get to the "second" Aperture Science Lab, the one where you use the propulsion gel.

    EDIT:
    ***Spoilers***

Actually there is a chance of accidentally skipping this. It happened to me yesterday -- there is a way for you to get to the elevator without needing to activate the bridge (and find GlaDOS). I noticed something was wrong when I started the next chapter and I suddenly had her in my possession.

Here is a video showing how you can skip this part (and not get the achievement):

Thanks to DarthMike431 for clearing this up, I wasn't aware of this.

P.S.: if you're going to rate down, please specify so I can edit the solution

SpyforhireYou can totally miss this achievement. I missed it not knowing what was going on. You need to edit your solution.

skronchi too completely skipped this and was surprised to see a potato on my portal gun. seems like more of an error on valve's level design than anything else

Gen Fire DragonDamn you people who missed it. You're only supposed to think with portals when you're supposed to.

Frankie DrumsNYMissed it too. Does this mean I could be going through these levels carrying Glados?

Torhu HybridNot sure why this is downvoted, but gave it an upvote because I didn't realize it was skippable. They almost should've accounted for this in the design...perhaps by having the lift sealed or something. Hmmm....

Playing through the whole game without GLaDOS would be an interesting bonus, make for an alternate ending or something. Missed opportunity LOL.

OnzaI almost missed it too. I did not read this first, as I was just playing through... But I failed to get to the control room. I was trying all sorts of stuff, and came close (similar to video), but did not quite do... Good thing otherwise I would have been VERY confused.
