Just need to kill 500 monsters for this achievement, and most will get it playing naturally through the game (although I didn't). Your companion kills count as long as they're in your group, and the kills are saved in an internal counter so you can save, kill a bunch of monsters and then reload and have them all count. One thing to remember is that this is not TOTAL kills but MONSTER kills, and there is some confusion on what constitutes a monster in Risen 2. This is a list of what is known to count/not count:

DOES count: Sea Creatures (Sand Devils, Giant Crabs, Sunken Ones, Leviathans), Giant Bugs (Termite Workers, Termite Warriors, Grave Spiders), Undead (Ghouls, Warriors, Guardians), Flyers (Cave Bats, Firebirds, Gargoyles)
PROBABLY count: Bosses (Earth Titan, Kraken, Gnome-Eater, Mara)
MIGHT count: Animals (Monkeys, Jaguars, Panthers, Warthogs, Alligators, Gorillas)
DON'T count: Humans (Pirates, Savages, any named NPC), Vermin (Rats, Turkeys, small Monkeys, Tamed Warthogs)

I personally don't think animals count for this achievement, but enough people have claimed to unlock it from killing one so I'm not entirely sure (my achievement popped after killing a Giant Crab for the record). My advice is to concentrate on what we know to count as monsters and here are a few good places where they can be found:
Isle of Thieves (~55 monster kills available)
Storm Island (~45 monster kills available)
Isle of the Dead (-45 monster kills available)
Termite Cave on southside of Tacarigua (13 quick termite kills)
Water Temple Entrance (6 quick sea creature kills)
Water Temple Final Battle (Leave her alone and she will continually summon waves of sea creatures)

During the last fight of the game Mara will summon enemies endlessly if you don't kill her! So you can just grind Glory points or KILLS if you want. By this method you can play as you like and in the end get class specific achievements.

There might be faster way to kill enemies because after killing enemy horde it will take about 20sec before Mara will summon a new one. But if you are like me and you have to grind some XP this method will bring you kills too quite nicely.

I had around ~470 kills after I finished the game first time before loading the game and killing enemies that Mara will summon. Despite I cleared all the different islands and explored them pretty well.

SOLUTION: During the fight don't kill her or attack her, she will eventually summon enemy horde. After you clear a wave just run away for about 20sec. Mara will chase you during this time but she will give up and summon more enemies. You need some provisions, painkillers and so on, but because this is the last fight of the game you probably have advanced character. If you have basic blade skills like parry, counterattack etc you will be fine. Always kill sand devils first, then Leviathan and finally Sunken Ones.

I got around 40k of Glory points and I had only Blademaster achievement (i could buy levels and reload the game). I had level 7 blades, level 6 cunning, voodoo level 4, toughness level 3 and firearms level 2. It took me less than hour to get required 20k of Glory points so after I got 60k of XP I bought required levels and reload the game and bought levels again and got cheevos.
