I got 1000 kills doing Chapter Challenge on Normal with Momiji (working towards my Kunoichi achievement) without even touching Trials. It unlocked at Day 6 or 7 and I was just going through the game as usual, skipping a few encounters in between. This should unlock naturally as you progress towards unlocking costumes 3-4 anyway, as you need to complete 25 & 50 Trials.
