  Once you pick the Hulk as a playable character, holding the (Y) button changes your character back and forth between Bruce Banner and The Hulk. Things can be sped up if you also enable the mini characters extra, which has the side benefit of making the transformation go more quickly.

  Any idea what the mini characters extra is unlocked please?

  This video (at 0:25) shows the location of the Mini Characters Red Brick as well as demonstration exactly what the brick does. The Deadpool bonus mission where you collect this Red Brick is called 'Bro-tunheim' and it is located at the northwest docks, but can only be accessed and entered when you have 200 Gold Bricks. This is the 11th and final Deadpool bonus mission in the game.

  Thanks KrazyK70! I popped this in like 5 minutes with mini characters enabled. Took me only 26 transformations though as I have almost completed the game so I had to have already transformed 24 or so times.

  If you left this achievement until one of the last like I did then load up the first level on 'story' mode (This is so when you hold down Y it doesn't go onto the character selection screen) turn on the mini characters extra as suggested by Toddard, use hulk as a playable character and then hold Y, after 5 -10 minutes, if that, the achievement will be yours

  Hulk is introduced in the first level, "Sand Central Station". All you need to do is press and hold the button and you will transform into Bruce Banner. Do not let go, but instead just keep holding it and you will continue to transform back and forth between the Hulk and Dr. Banner until the achievement pops. You can use a rubber band or something heavy to hold the button down for you so that you can go do something else and let the game do it for you.
  
  You can also just wait and play through the game as you will have other opportunities to transform back and forth for puzzle solving as you play through levels.

  This is the guide here out of all of them. Right at the start of the game just hold Y, simple. Takes maybe 10 minutes.
