This achievement is very similar to the Il Principe achievement in AC: Brotherhood, however, it is much simpler. For this achievement, you have to obtain a 100% synch in all main story missions.
It is recommended that you just complete and obtain 100% synch as you go, rather than simply completing the missions and revisiting them later - it'll save a lot of headache later on. If you are in a mission and fail the optional requirement, then you will need to completely restart the memory, regardless of checkpoints gained. This can definitely be a pain, especially if you fail late on a very long mission.
Each main DNA Sequence varies in length, with some memories having optional objectives and some not having optional objectives and are more cinematic than anything.
