Getting 40 people is easiest to do on a difficulty of 100% or less, with realism mode turned off. Any types of characters count for the 40, and they can either be in an expedition or the city they will all count for the total.

In Thea your population grows in three ways.

1) Attract new characters randomly with buildings built with certain materials. Certain buildings come with a base level of attracting other humans. For example, cabbage patches and pastures are easily constructed and will have at least 1 attraction value for humans. The Manger is the same way but for beasts. Characters attracted this way will just show up in your city randomly.

2) Completing events can often reward you with a character. Many of these events can repeat through the game as well, if you complete them the same way you will often continue to get new characters.

3) Children growing up is the last way to gain characters. Often you'll gain the child resource though events in the game. They will sit in your inventory until they randomly grow up.

All of these 3 types of events has a chance to occur on any turn, the percentage chance varies based on your current population and the turn #. For example, if you have a population of 30+ around turn 200 the percentage chance for gaining new characters will be very low. As you continue the game through turns 300+ the chance will increase until you reach the new expected population level. By turns 500+ having 50 or more people is not uncommon.

You should also avoid losing characters as you pursue this. If a random event kills off a bunch of your child population you may want to reload your save. Have a medic in your party and attempt to avoid losing them due to wounds.
