You will need to have 4 controllers for this achievement, as there is no online multiplayer.

I would HIGHLY recommend unlocking the character Payne and using him, as he is probably the best of what the game has to offer.

Please note that use of Nullifying cores ('Pull' and 'Collateral Damage') will disable the achievement from working. The strategy therefor would be able to start with no cores activated.

The good news is, you can make this achievement a little easier by starting single player and adding/dropping players as required to make the progress more manageable.

To elaborate: If you start the game with four players (or more than one in general) you each have 1 life and the ability for unlimited respawns. HOWEVER, respawns only work if there is a character sill alive on the screen. If all players are in process of respawning (takes about 10 seconds), the game will end. Also, the more players active at any given time make it so there are more enemies spawning. If you start a multi-player game and have the rest of the players drop out (Press Start -> Drop out {can only be done if you are still alive}), you will esentially be in single player mode, but you will only have 1 life.

Now, if you started the game in SINGLE player then had the other players join in, the premises are the same except when other players drop out and you are back to 'single player' mode, you will still have 4 lives (default for single player mode).

So, start a game in single player mode and play through with only the one player (this will be much easier, as there are fewer enemies with only a single player). If you want to verify that you not wasting your time, you can clear out any room and instead of exiting, have the other controllers join the game. Immediately have the other 3 drop back out and check your Xbox Achievement progress for this achievement. The progress bar should reflect based on how far you are towards room 50.

Anyways, you will just need to play single player all the way until room 49 and then have the other 3 controllers join in before making your way to the exit. When you enter room 50 with all controllers activated, the achievement will unlock.
