This achievement is unlocked by landing the killing blow with a fully charged, upgraded Mega Buster shot on a boss. You have to first unlock the Arm Upgrade from Flame Mammoth's stage, or if you did not get the arm upgrade, you automatically receive it when defeating Vile on the first Sigma level (see Re-Armed achievement). I recommend whittling the boss' health down using the weapon they're weak against first to make things easier.
