500 relics..a little bit of a grind, but not bad.
Every expedition has 3 parts or floors to the expedition for a total of 9 relics.
Each floor will track relics per part. 33/66/100%. that way you can easily keep track so see if you have them all.
Only use the portal to the next part once you have all 3 relics on that section.

Relics can be tracked in Journal > Achievements > Quests > Relic Hunter (I, II, II)

Finishing off the campaign expedition quests before you unlock the dailies will give you 45/500 relics.
Once you complete the Master Expedition quest you will be able to unlock the expedition dailies, which can be repeated 3 times a day. (27 relics a day)
This should take around 17 days of dailies (assuming you miss no relics)
Tested - you can do as many as you want each day (by helping other), but you can only get 27 relics a day from dailies.
Jappe confirmed you can not hold onto the quest, do more runs and get additional relics. (Thanks for testing this theory Jappa)

Updated note : If you are in a party, it doesn't matter who collects them as it will still count for you. (but only on your first 3 dailies, not after that)
Update note number two : Seems like the maps with the bridge and lava, can sometimes bug out only giving you 8 for the run (3rd piece cant be found) - not always the case though. Seems like the map with the bridge there is one close to the camp fire at the start - but it blends in and give people issues - double check this one otherwise its a lot of running back
    (Thanks Jappe)

    Happy hunting.

Shadow 00 FoxJust a note, don't abandon the daily quest, there's this note in the quest description: "Warning! Dropping this mission will count toward the repeatable number." lol

JappeThe map with bridge and lava doesn't have a bug and the third relic is there. It's behind where you spawn. You need to jump over the stone obstacle and it's behind that. Not sure if it's intended or not but certainly not a bug.

Shadow 00 FoxI already gave up getting these... it takes a long time and the last time we had a goblin glitch into a mushroom and aggro us when we tried to pick up the relic nearby. Ugh.

OnsidicYeah, one being seen recently is the lava bridges enemy not found.. and at the end of a floor portal, enemy gets stuck in it.
Sometmes everyone dieing works, sometime randomly slashing close by also works

JappeMost of the time if everyone defeats themselves it fixes itself. I've been doing the MEs trying to get the Alabaster weapons and I'm already at 384 relics collected and I've even missed some and failed a quest once by going out of the ME thinking I could go back in. There's a lot worse grinds in this game than this one lol

JappeOh and I tested yesterday that I did NOT get any additional relics to the achievement tracker if I didn't return the quest and still went in a 4th time with others.

Fuzzmeister JIf you abandon the rune-etched armor quest, you can't put down runes at the start or get a reward from the chest at the end. You can still work on this achievement, but it's a little less rewarding if you don't keep the quest. You can just pick the quest back up if you already got rid of it.

OnsidicThanks for testing my theory Jappa - added update to main. Yeah free loot grind, more like running an easy dungeon that the grind that other campaigns have
