  You must have the Spider Rune to access this area. Once you've acquired this, on your next run take the following path:
  

  1. Prisoner's Quarters
  2. Toxic Sewers
  3. Corrupted Prison

  The game let me access this area even without purchasing the Bad Seed DLC which is annoying because now I've unlocked an achievement in a DLC I am now pressured into buying (because I'm an annoying completionist, ha)

  I assume this should unlock immediately upon entering Corrupted Prison, but it didn't. Any advice for triggering this?

  It's been a while since I've played the game, but I do think it should unlock immediately. You do have to go through the door from the "Passage" between biomes for it to unlock. If it didn't unlock the first time, you might just need to play through again.
