  You first get introduced to Poppy during the main story quest "Beast Class" - shortly after that you'll start progressing her relationship questline. There's a total of 9 quests to complete for her relationship questline:
  

  1. Fire and Vice (Main Quest)
  2. A Dragon Debrief (Side Quest)
  3. Poached Egg (Side Quest)
  4. The Poacher's House Call (Side Quest)
  5. Surprise Meeting (Side Quest)
  6. The Centaur and the Stone (Side Quest)
  7. It's in the Stars (Side Quest)
  8. A Bird in the Hand (Side Quest)
  9. Poppy Blooms (Side Quest)

  Note: In most cases you'll need to advance the main story questline before her next relationship side quest will appear.

  This was glitched for me the first the I talked to her, but it popped after loading an autosave just before it and speaking to her again

  Glitched for me. Didn't unlock after speaking to Poppy in great hall for poppy blooms. Tried loading other saves, resetting game all to no avail. Very annoying.
