This is done as part of the story. After you finish DNA Sequence 3, as part of your mission for DNA Sequence 4 (Don't want to give away the story) you will go through some catacombs that lead to the Assassin's Tomb under Santa Maria Novella. When you exit the Tomb the achievement will pop.
