This is an achievement for the Peggle Alums. It won't be as hard as Doctor of the Peggle Arts, but its still makes Peggle Professor look like a cake walk.
First of all, there are 60 new challenges for your peggling pleasure. One of the things that makes these levels harder is all the moving parts, which most of the levels have. Also, on top of the usual challenges, they throw a couple of new curveballs your way. Since there is no sure fire way to beat Peggle besides just playing through it, stepping your game up and hoping for a tad bit of luck, I'll just be presenting a couple of pointers for the different kinds of challenges.
For most levels I use Warren (Lucky Spin) as my default because he can get the most points. If you are going strictly for eliminating pegs and completing levels, then Master Hu (Zen Ball), Splork (Space Blast) or Jimmy Lightning (Multi Ball) are your best bet. It is a good rule of thumb to use whichever character the level is themed after if you're not having much luck.
Another key skill is learning how to get the ball back into the bucket for easy free balls. There are so many times when you can barely survive a level with one ball simply by banking it off one peg and straight into the bucket. It is slow, but it is an easy way to get points and it'll keep you afloat on some of the harder challenges.
The last major tip for any kind of level you are stuck on is to check the leaderboards. You can look up the top scores for every level in the game and you can see which character those people used. Looking up the Best Shots can also be very useful when you're going for major point challenges.
1-1 through 3-5: Use Warren and if he fails use the other 3 recommended masters.
4-1 through 4-5: Purple pegs are your friends here. Make use of them for maximum points.
5-1: Long shots, double long shots, super long shots. Master these and you'll do fine. Off The Wall is another easy one. Kat Tut (Pyramid) is also good because his bonuses are the easiest to get: Pyramid Pick-up and Eye of the Pyramid.
5-2: Bjorn (Super Guide) is great for this. About time he was useful.
5-3: Once again long shots are your friend and you can get 3 different kinds. Kat Tut has two easy style shots in Pyramid Pick-up and Eye of the Pyramid. Also, if you hit both green pegs in one shot there is another one for you. Off The Wall is easy to get. Free Ball Skill is also easy to do. You can get Orange Attack, Mad Skillz, or Extreme Slide in there with enough practice.

5-4: Purple Pegs and Free Ball Skills. Restart if the orange peg placement isn't to your liking.
5-5: Free Ball Skills are key here. I recommend Warren to get extra free balls.
6-1 through 7-5: If you did the challenges from the main game then you pretty much know what to expect here. The AI will make the most insane shots possible. The key here is to make it hard for them to get the opportunity to get those shots. For example, try to get the green pegs before they do and never ever let them get the final shot for Fever. Also, don't shoot without hitting an orange peg because your score will go down 25% every time.
8-1 through 8-5: Your best bet is to go with the characters that are tied to the levels, but Master Hu and Warren are the two best alternatives.
9-1 through 9-5: To me, these are the worst ever. Some of these levels are hard to complete, let alone score a high amount of points in. Pace yourself. If you don't get a certain amount of points by the end of the first level you may as well start over. If Warren's Triple Score isn't helping you, then try going for as many style shots as you can. Don't depend on getting the 100,000 fever jackpot because 9 out of 10 times you will be disappointed. Check the leaderboards to see which masters the Pros used to get their ridiculously high scores.
for 9-5 specifically: I used Warren, Master Hu, then Warren again. I didn't bother moving to the second level unless I had at least 250,000. With luck you'll be at half a million by the end of the second level. The third is the real money maker. I used Warren and worked on getting a Triple Score ball inside the loop in the middle so that it hits every peg. This will get you major points almost every time especially if there's a purple peg in there.

10-1 through 10-3: These levels are just plain fun yet frustrating. They are very dependent on orange peg placement so keep restarting until you see a formation you like.
10-4 through 10-5: These levels go against everything you've learned up until now. Use Bjorn since his power doesn't actually net you any involuntary points. Make sure you finish the level with your last ball. If you have extras just keep firing them straight to the bottom before you hit that last peg. Try your best to avoid style shots and just hope you make it into the 10,000 fever pot because ironically enough, that ball became very fond of the 100,000 pot when I tried those particular challenges.
11-1 through 12-3: Once again your Free Ball Skills need to be at their peak for the later levels. Use Warren since you're guaranteed an Extra Ball if nothing else most of the time. Restart to find the easiest looking orange peg placement.
12-4: This one is up to you. Once again, check the leaderboards for best shots and you can replay the top ones. You can even use levels from the main game and not just Peggle Nights. Most people would probably use Stage 11-5 because that's the most commonly used one from the exact same challenge for the main game. I actually used Nights Stage 3-2 with Warren. It is very easy to angle your first shot so that you can get two Extreme Slides. If you can get Triple Score and Magic Hat in the mix, then you're as good as gold.
12-5: This one isn't as daunting as it seems. If you've done everything else, then this should be a breeze. Follow the same steps for 8-1 through 8-5.
