Like the following achievements, this can be done on Lagoon City Zone Act 1, with the boss battle.

The Heroic Blue Blur achievement in Sonic Superstars worth 55 pointsUsed Sonic's Drop Dash to defeat 100 enemies.

The Aerial Takedown Artist achievement in Sonic Superstars worth 61 pointsUsed Tails' aerial attack to defeat 100 enemies.

The Crimson Guardian achievement in Sonic Superstars worth 60 pointsUsed Knuckles' glide to defeat 100 enemies.

The Tenderhearted Hammer-wielder achievement in Sonic Superstars worth 58 pointsUsed Amy's Hammer to defeat 100 enemies.

The Secret Achievement achievement in Sonic Superstars worth 65 pointsContinue playing to unlock this secret achievement

This boss spawns pond skaters enemies, and if you hit the boss more than 2 times, it will stop spawning enemies.

The achievement unlocks at the moment of defeating the 1000th enemy

I recommend trying to unlock the other achievements first, and this will unlock without grinding (eventually)

Do you need to complete level without dying for kills to count?

I'm pretty sure that is not required. While boosting for this as Knuckles, I died at least 2 times and I keep my kills.

Also, forgot to mention it, every achievement related to defeat enemies unlock at the moment of defeating the 100th or 1000th enemy
