The boss is Gretel who Nier originally fought along with Hansel. Gretel retreats to the Lost Shrine. Through normal story progression, Nier encounters Gretel again and fights a final battle. The battle is in three stages. In the first stage, Nier must defeat numerous shades. Until he does so, Gretel is invulnerable. Then Nier fights Gretel twice. A story related cut scene occurs between the two fights. After the second fight, Nier defeats Gretel once and for all and, if the time goal of two and a half minutes has been met, the achievement will unlock.

Gretel is not terribly difficult to defeat. There is a save place before the battle so it can be repeated if the time goal is not met. The issue is that the cut scenes count against the time so they need to be skipped to facilitate getting the achievement. On the other hand, they contain relevant story information so you will probably want to watch them the first time.

The fight recurs for each walkthrough--working towards the A, B, C, and D endings. Nier will be progressively stronger during each walkthrough until he reaches max strength. So, depending on how fast you have been leveling up--a personal choice--you may want to defer getting this achievement for a later playthrough when Nier is at a level where he can quickly defeat Gretel. On the other hand, if you have leveled up Nier rapidly, the achievement can readily be obtained in a first playthrough.
