Some sites explain that there is a way to get off to a good start on MGS Phantom Pain by downloading saved data from MGS Ground Zeroes.
One of them is ripping all the prisoners, two targets that besides being eliminated must be extracted, and two agents that are disguised.
But my question is this:
After I rescue all the prisoners, the two targets that would have to be eliminated and the other two agents disguised, can I complete the difficult way without rescuing any prisoners, and killing the other targets?

Yes. The way I do it is easy and it virtually guarantees an S ranking. It's easier with reflex on and you have enough wriggle room that the penalty will not keep you from an S ranking.

When you start out, there is one enemy on look up go right, sneak behind him, and take him out with the stun pistol. Jump down to the ridge and go low at the end and take out the guard there. Turn right up the road. If you want, get some extra stun rounds in the empty guard shack. Go low just before you get to the guard shack. Stay low turn left hugging the wall on your left until you get behind the watch tower. Shoot the guard in the watch tower.

Now you can run until you get to the fence. Crouch and hug the fence until you get to the end of it. One of the targets is walking with two escorts. Shoot all three. This is where reflex really helps. Shoot the escort to the rear. The target and the other escort will notice. With reflex on, it is very easy to stun them both with head shots.

Now, duck low and crawl left until you see the guard up in the tower. Stun him. Go back and pick up the target. He is bald and the only one with no hat. Pick him up. Crouch and go cautiously left of the truck until you can see a guard. Stun him. To the right of the guard you just stunned is a jeep. Put the target in the jeep. Now, crouch up the hill then crawl through the gate in the chain fence. This is a check point.

Once inside the fence, crawl left. Towards the end is a guard in front of a red door. Mindful that there is no other guard in the area (if one is close let him walk away) stun the guard while in the crawl position. Crouch, pick him up and take him inside the red door. Crouch along then crawl until you see a guard. Stun him. To the right is a ladder leading up to a walkway that takes you to the big control tower. That's where the other target is with two guards.

Go up the ladder, crouch, then crawl. You will be able to see an escort. Stun him and crawl a little closer. The target will come into view and investigate the stunned escort. Stun him, too, from your position prone on the walkway. The second escort will now investigate coming into view. Stun him, too.

Now, proceed cautiously up the ladder to the control tower. On hard, you might be detected if you are not crouched. Pick up the target. He is the only one without a hat. Return the way you came. When you exit the red door stay crouching. You previously took out the sentry so the way should be clear but there is a slight possibility you will be seen on Hard if not crouching. Hug the fence on the right until you get to the gate you used to enter. Keep crouching and go to the jeep. Put the target in the jeep. Get in the jeep and drive it straight until forced to turn. Turn right and drive straight. This will take you out of the mission. You will have extracted both targets in a very fast time, no alerts, no kills, for a comfortable S ranking.

There is a cassette tape in an armory close to where you took out the first target. I recommend picking it up on the playthrough where you are required to mark all targets.

I understood how to do the mission in the S rank, but I would like to know if it is necessary to rescue the targets in both difficulties, or if I can only rescue in the normal difficulty and kills them in the difficult difficulty.

Just rescue in normal, mark the targets in normal, & play anyway you want twice in Hard. This gets you your S rank & unlocks all trials. There is a cassette tape that you need to get on any one of the four runs. It is in the armory near to where the first target starts off.

Many thanks for the advice and tips, I had already completed Ground Zeroes 100% once, I achieved S rank in all missions, both normal and difficult. What I did not know, was that rescue all the prisoners and the other four characters, they would help at the beginning of Phantom Pain ..
So I asked for help because I did not know the way it had to be done ..
He was in doubt whether he could rescue everyone in the normal difficulty and kill them on the difficult difficulty, or if he would have to make the rescue on both difficulty levels.

I played Ground Zeroes & Phantom Pain on the PS4 when they first came out. There are a number of bonuses. If you rescue these two targets (and you only have to do it once), you get special volunteers at the Mother Base. One of them is Kojima. I assume you get him when you extract the intel operative. I believe that every single prisoner you rescue adds to the number of volunteers. IIRC, every mission has at least one prisoner even if there is nothing about him in the mission statement. When I was doing the part where you had to mark all targets in each mission, I went ahead and made sure I identified and rescued all friendlies.

I just picked up Metal Gear Solid V: The Phantom Pain on GwG. Since Metal Gear Solid V: Ground Zeroes was on sale for, I think, 75% off; I went ahead and got it, too, as well as the 360 version. My understanding is that you only need one save file. Either the 360 or X1 Ground Zeroes save file can be uploaded and will work with both the X1 & 360 versions of Phantom Pain. Just to be safe, I played both.
